Community Practice Grand Rounds: A Multidisciplinary Framework for Integrating HER2- and TROP-2–Directed ADCs Into Personalized Management of NSCLC

There are multiple advances in the personalized way healthcare professionals can treat non-small-cell lung cancer (NSCLC), depending on tumor genetics and the patient’s response to therapy. When patients are diagnosed with NSCLC and are predicted to respond to HER2 or TROP2 therapy, systemic treatment with antibody–drug conjugates can be tailored to improve outcomes. This presentation will highlight these changes, providing clinical trial evidence and key points for patient management, focusing on HER2 and TROP2 options.

CME/CE Info

Goal Statement
The goal of this activity is to improve the knowledge, confidence, and competence of learners to integrate HER2-directed and TROP-2–directed ADCs into optimal management of patients with NSCLC.

Target Audience
This activity is intended for community medical oncologists, nurse practitioners, physician associates, nurses, pharmacists, and other healthcare professionals caring for patients with NSCLC.

Learning Objectives
Upon completion of this activity, participants should be able to:

  • Differentiate HER2 mutation, HER2 tyrosine kinase domain mutation, HER2 IHC 3+ overexpression, EGFR mutation status, and investigational TROP2-NMR/QCS biomarker status to guide appropriate selection of approved or investigational HER2- and TROP-2–directed ADC strategies in NSCLC

  • Select and sequence HER2-directed and TROP-2–directed ADCs for appropriate patients with NSCLC, with consideration for prior therapy, biomarker status, CNS involvement, performance status, comorbidities, toxicity risks, patient goals, guideline recommendations, and clinical-trial availability

  • Implement multidisciplinary workflows to prevent, recognize, grade, and manage key ADC-associated adverse events, including ILD/pneumonitis, stomatitis/oral mucositis, ocular toxicity, nausea/vomiting, and myelosuppression

  • Identify patients with NSCLC who should be considered for clinical trials evaluating HER2- or TROP-2–directed ADCs, ADC combinations, and biomarker-selected strategies, and apply practical approaches to referral from community practice
