Conference to Clinic: A Test-Your-Knowledge Challenge of Emerging Next-Generation KRAS G12C-Targeted Therapies in NSCLC

This activity reviews evolving evidence for targeting KRAS G12C in non-small-cell lung cancer (NSCLC), with emphasis on the clinical limitations of first-generation KRAS G12C inhibitors and the rationale for next-generation agents and combination strategies. Faculty discuss the biologic role of KRAS mutations in NSCLC, mechanisms of intrinsic and acquired resistance, and emerging clinical trial data evaluating KRAS G12C inhibitors with immunotherapy, chemotherapy, and perioperative approaches. Key considerations for learner decision-making include efficacy outcomes, toxicity profiles, PD-L1 status, comutations such as STK11 and KEAP1, prior KRAS inhibitor exposure, and how these factors may shape treatment selection, sequencing, and clinical trial discussions for patients with KRAS G12C–mutated NSCLC.

This activity is intended for oncologists and other healthcare professionals who care for patients with non-small-cell lung cancer. 

Program Learning Goal

This activity aims to improve learners’ knowledge, confidence, and competence in integrating novel targeted agents to improve outcomes in patients with KRAS G12C–mutated non-small-cell lung cancer.

Learning Objectives

Upon completion of this activity, participants should be able to:

  • Identify patients with NSCLC who may be eligible for enrollment on clinical trials with next-generation KRAS G12C–targeted therapies

  • Evaluate how next-generation KRAS G12C–targeted agents in late-phase clinical trials might be integrated into real-world clinical practice for patients with NSCLC

  • Discuss with colleagues and patients ongoing advances in targeting KRAS G12C for solid tumors
