In this second of 3 video modules, an expert reviews emerging clinical evidence on CDH6-directed ADCs for the treatment of ovarian cancer, with a focus on platinum-resistant disease. The faculty examines CDH6 expression and its relationship to FRα expression, along with early-phase efficacy and safety data for investigational agents, including CUSP06 and raludotatug deruxtecan. Findings from the CUSP06-1001 and REJOICE-Ovarian01 trials, including objective response rates, response durability, activity following prior ADC treatment, dose optimization, and adverse event profiles, are highlighted. Clinical outcomes are also compared with those of other ADCs evaluated in ovarian cancer, providing context for assessing the potential role of CDH6-directed therapies and interpreting emerging evidence that may inform future treatment selection and sequencing.

Efficacy and Safety for CDH6 ADCs

Pre Assessment

Assess your current knowledge and clinical approach before beginning your video.
1.

How many people with ovarian cancer do you provide care for in a typical month?

2.

For those who practice in academic or community settings, please indicate your practice setting:

3.

You are treating a patient with ovarian cancer who has failed 1L with platinum and taxane therapy. Cell membrane staining of a tumor sample shows CDH6-positive ≥1% (1+/2+/3+), FRα-positive (≥75% of viable tumor cells with 2+/3+), and HER2 (1+) scoring results.

What FDA-approved therapy would be appropriate for this patient?

4.

The phase II/III REJOICE-Ovarian01 study reported which of the following outcomes for patients with high-grade serous or endometrioid ovarian cancer, primary peritoneal cancer, or fallopian tube cancer treated with R-DXd?
