Precision Diagnosis and Disease Biology of TGCT—Establishing the Rationale for CSF1R Targeting

This downloadable slide deck reviews the clinical presentation, diagnosis, disease biology, and multidisciplinary management of tenosynovial giant cell tumor (TGCT), a rare, benign but locally aggressive synovial neoplasm that can cause substantial pain, swelling, stiffness, recurrent effusions, and functional impairment. The activity distinguishes localized from diffuse-type TGCT, including differences in growth patterns, recurrence risk, and the limitations of surgery in infiltrative disease. It also examines the roles of MRI, CT, and pathology in confirming the diagnosis, defining disease extent, identifying bone involvement, and excluding malignant mimics. In addition, the slides show how CSF1 overexpression and recruitment of CSF1R-expressing inflammatory cells contribute to tumor growth and symptom burden, providing the biologic rationale for CSF1R-directed therapy. These concepts are important for improving early recognition, referral to experienced multidisciplinary teams, and individualized treatment decisions that balance disease control, procedural morbidity, symptom relief, and preservation of joint function.
